that..Brenda was her character from Beverly Hills 90210 during the shows prime. I'm not sure what started it (I THINK it was Brenda's relationship with Luke Perry's character but I never watched the show so I'm only guessing) but, in America at least, there was a huge "I hate Brenda" campaign going on against the character and by extension Shannen Doherty.
So Rene's offended that Willam has mistaken her for Brenda because, I guess, she hates Brenda too.
I don't think there's really a "purpose to the joke" as you put it, I think, more than anything, it was a way for Kevin and Co to fuck with Ethan Suplee (Who they told that Shannen didn't know he was going to say it so he thought her reaction was genuine.)
I don't know, its so silly and out of nowhere that it makes me laugh regardless.
